A clinical trial has found that selinexor, the first of a new class of anti-cancer drugs, was able to shrink tumors in almost a third of patients with recurrent glioblastoma, an aggressive brain cancer.
Glioblastoma is an incurable brain cancer that needs new therapeutic approaches. Considering that few treatments have any measurable effect on recurrent glioblastomas, the results are encouraging, says the studys leader, Andrew B. Lassman, MD, the John Harris Associate Professor of Neurology at Columbia University Vagelos College of Physicians and Surgeons and chief of the Neuro-Oncology Division at Columbia University Irving Medical Center/NewYork-Presbyterian.
The drug induces a response in certain patients, and several trial patients stayed on selinexor for more than 12 months, including one for over 42 months, adds Dr. Lassman, who also is associate director for clinical trials at the Herbert Irving Comprehensive Cancer Center.
The study was published January 10, 2022 in Clinical Cancer Research.
Glioblastomas are typically treated with a combination of surgery, radiation therapy, chemotherapy, and sometimes an electrical device, but the average survival time is just 12 to 18 months.

Natural tear lubricant was applied to the eyes. For each mouse, 5 × 106 U87MG cells in 5 µL serum-free Dulbeccos Modified Eagle Medium were injected into the right hippocampus using a 10 µL Hamilton syringe at the following coordinates related to the bregma: 1.5 mm lateral, 1.5 mm posterior, and 3.5 mm in depth. The needle was lowered to 3.5 mm and retracted by 1 mm, before injection . Instruments to control rectal and cranial temperatures were closely maintained at 3637°C before, during, and after the procedure. The incision was sutured using sterile black monofilament nylon 5.0, and the area was cleaned with betadine. Mice were individually caged, observed daily for body weight, temperature, and locomotor changes. Animals were perfused at the end of the 30-day survival and brains removed for ex vivo MRI. If youre able to become pregnant, you should use effective birth control while youre receiving Avastin. You should continue to use birth control for 6 months after your last dose.

Temozolomide and bevacizumab are two popular chemotherapeutics used to treat glioma.

- Temozolomide works by inhibiting the growth of rapidly dividing cells. This medication binds to an area on the DNA of cancer cells, causing them to self-destruct. Some tumors, such as certain mutations in glioblastoma, may have mutations that make them more susceptible to temozolomide treatment. Of note, temozolomide is often taken by mouth, in pill form.
- Avastin stops blood vessels from growing. The idea is to prevent tumors from creating new blood supply, thereby cutting off the blood supply to a glioma and preventing them from growing or recurring. This medication is often administered through an IV, directly into the bloodstream.

Avastin is FDA-approved to treat metastatic kidney cancer. Metastatic kidney cancer has spread out of your kidney into other parts of your body.
For this use, Avastin is given in combination with a drug called interferon alfa.
Effectiveness for kidney cancer
In a clinical study, researchers looked to see if using Avastin with interferon alfa was more effective in treating kidney cancer than using interferon alfa with a placebo.
The study showed that people taking Avastin with interferon alfa had better overall outcomes than did people taking interferon alfa and the placebo. In particular, half of the people using Avastin with interferon alfa lived for at least 10.2 months without their cancer worsening.
